I've never seen a stone in liquid culture, let alone this!

A friend gave me a mycelia syringe and I neglected it for monthes. I pick it up and find this:Upload

I was like.....whoa.
Now how do I get that outta there?

I hope I can get some tissue out of there for a culture....jalisco doesn't normally get stoned like that....must be a good one :-)

uheemm why don't you clean up get it inside of a flowhood and slice open the syringe?? from there treat as you would clone tissue......

You don't have to slice open the syringe, do you?

Can't you just pull the plunger out of it very carefully in a glove-box or flowhood?

Thats the ticket....

But I can tell it's going to be difficult. I think I will sterilize some air in a jar, insert the needle, and then pull the plunger....also in front of the hood.

If only I had a biopsy needle......then I could extract a sample from the center of the stone without doing anything but unscrewing the needle.

Just open it up.....when stone forms, it has a protective covering, it will not contam......altho I admit I've never had experience with "Hydro-stones".

You should pull out the plunger and then mix a soln. of 1 part 3% peroxide to 9 parts water and then just blend the soln. with a blender or some type of mixer to break it up. Then, use that for a syringe. If you do all this in a glovebox you should be able to clone it pretty simply.

I'm not gonna blend the whole stone.....

I'll try to extract a culture from it, piece by piece...so I don't lose it....

eventually I'll get a clean plate
if I use peroxide agar

but for now it stays in the fridge....no lab right now until I move.
