I think this story is probably an urban myth, but my ex claimed that when he was in HS bio class, everyone scraped their cheek cells for staining and microscopy (good thing to add to your catalogue, cat).

One student raised her hand and asked what the tadpole looking things in her cheek cell slide were.

Nope, malabar coast, they look like pans though. I suspect that's because there's not enough moisture for normal fruit formation (size).

Sooooo, it's been awhile. I am not going to lie, I've let my agar work fall WAY behind. As usual though, the mycelium has surprised me again. Pictured is self isolating rhizomorphic growth of a malabar culture that has actually run over already colonized agar. I will transfer this to continue the isolation.

Not pictured, the Hawaiian pans that went from "milked" agar to oats attachicon.gifpeacefrog.gif.

Sorry for bumping and posting some pic from ur old thread brother cats.
Found this from my old petris. While checking on your informative threads.

It Shows some resemblance to ur culture.
Rhizho crawling & growing back on top of tementose.
She`s unique and only 1 out of 20 old plates.
Sorry for my crappy camera.

Will try to TWEEZER some rhizomorphs out from it.

~ Did you grab those long mycelium sectors from 1-3 oclock?

Just knocked up few tea agar out of it.

I also loved how cubes grow on dfa. After i switched from brf(agar) 6 months ago.
Just need to clean my culture from DFA bacteria.
Would love to try ur catitives and recipes on 2nd transfer,
Hope i can train/trial some for chitin digestion someday, and put this plenty of stock of mine in use.

You also mentioned that you dont flame anymore.
I also want to cut back from flame sterilizing. Esp. After i got a clean culture.
I also feel that needles are better sterilized in the pc instead & no flaming- based on boyle's law & thermo dynamics of air suction.

Since pftek standard has no SAB/PC which requires glowing needle at the expense of air suction & air spore load.

I did no flame on half of the plates on the picture to test. (Just a tool dip in paracetic followed by submerging in 75% ethanol) problem is 75% doesnt evaporate that quick inside the high humidity SAB. I should have sourced some 90% first.
Also forgot that i have those uv-c wands laying around,
Im thinkering that i may put it in use nexttime to let the etha evaporate while the tool is under irridiation.

I buy pre sterilized scalpels inoculation loops and have bags of needles.

I wrap my syringes in foil and throw them in my pc when I do short runs like agar.

I'll respond more when I'm on my laptop. I have a couple agar fails to post.

Word to the wise, adjust your agar recipe if one uses corn syrup as the dextrose source (or use dry dextrose) or the above will happen. I've actually grown myc on sloppy agar like that before but it's not ideal for obvious reasons. :biggrin:
