Agar Inoculation Noob Concerns

Opened up my 1st batch of agar inoculates thinking they were or were close to ready for transfer... they don’t so good

Serious bacteria issues. Did you use a syringe or a print? I am sorry, but they all look bad to me. What was your process?

One is right.

I have been plagued with bacteria problems with agar plates.

If all of them contam, it is most likely the syringe, or environmental inoculation issues.

Aww my heart just sank w/ that but I appreciate very much your response. I followed a potato dextrose agar recipe from McKenn’s “Growers Guide” which was as follows: 250g potatoes / 15g agar / 10g dextrose / 1.5g nutritional yeast. If they are all up the creek, it may have been my attempt to leave the jar lids loose during the pressure cooking sterilization portion? Second guess would be during inoculation, water from the caps/ creases falling down the sides of the jars? Not to say that the issue didn’t arise on my end but I was hours of meticulous using a makeshift SAS (Still Air Station), torch for reheating the syringe, and a whole family of cleaning agents So bummed about this batch... do you think it’s even worth attempting to utilize the leftover agar (kept stored in the fridge)? My scale at the time of making may have offered a few grams inaccuracy to the P.D.A. but perhaps that was another contributing factor? Again I appreciate your input and guidance as my first attempt at the psilocybin process AND experience thanks again all

That sounds like a lot of nutrition. You did make the full liter?

Also, that leaving the lid loose can be the culprit, especially if you forget to tighten them. (I've done that). Directions that say "leave the lid loose" are always vague. Here's how you do it, when you put the lid on, screw it down til snug, then back up 1/4 turn. Then don't forget to snug it when you take it out of the pc.

Some tips:
When using a recipe like that, Don't forget to adjust volume. Make your potatoes broth, add water to adjust the volume backup to 1000 ml. Bring a boil add other nutrients, when ready, check volume again, bring to a simmer and agar, as efficiently as you can.

After you pour the jars, let them cool open air to let out some condensation.

After your pc run, let them completely cool before opening the pc.

How much did you drop onto the agar? I have never had good luck with syringes to agar. Others have, I personally have not. If you are getting more than 1 drop on agar, it is too much. You would be better off doing an LC and going to agar with a mushroom clone. It sounds like you were clean enough. If you knock up a pf jar, or whatever you are using for spawn if not pf, you will know shortly if the syringe was a good one.
