Misfit, this is super cool. Congratulations on getting such a great position!

 

Does your lab look up protocols/media recipes for each species from different papers, do you have some reference you use for most stuff, or do you do a lot of trial and error attempts?

I can answer this a bit more in depth tomorrow. Trust me I asked about it. It’s a mix of research proven traits for certain species and also some current experiments.

So each type of fungus has a certain type of agar that is usually used except for experiments. I’ll get more info on basidiomycetes. I know that’s what everyone is interested in. I am also gonna see if we can start some Lions Mane or something.

No. In the lab. They have full trailers set up as fruiting chambers.

Got a little bit of Science going on today. Trying a few things for the first time. Had about 10 plates that seriously needed some attention. Moved several fruit bodies to new medium. Standard PDA. May get creative and try some V8 next batch. Only have 3 plates left anyway. Currently have African Transkei, Menace, and Huautla.
Another new method, to me at least, I took the left over Huautla scored the mycelium and running mycelium with distilled water in magnetic stirrer. This is going on oats today. Also starting one jar of Lions mane on brown rice this week.
Also running my last ACRB extraction will probably try a defat cause that shit looks like mustard seed.

I also rebuilt/ upgraded my PC if anyone is into that shit. Ryzen 3700 upgrade from intel.
I’ve been super busy but I miss you guys.

I've been prepping brown rice as described in Don Shadow's myceliated grain tek. Comes out perfect. I have zapotecorum and caerulescens growing out on it now.

Got more information on the V8 agar. None of us will be using it. It’s mainly used for ascomycetes. I also believe it is mainly used to force sporulation as fast as possible.
The standard agar used for basidiomycetes is a glucose yeast agar. So grab a pen. This is the one you guys would be interested in. For this recipe I use the following per 500ml. 5g glucose. 1.3g brewers yeast (LME works here right?) and 7.5g agar. Dextrose could be used in substitution here.

I’d say it should work just as good. Not sure though. It is just a different form of sugar though. Maybe I can try a small batch. And do side by sides. May be a week or more before I can though.

If you have time, I'd be curious about your results, It is essentially unrefined glucose. If you are ever bored, maybe give sorghum molasses a try too?

I hardly have time to run my own agar at home right now brother. Full time work and school and internship. Hahaha kill me now.
