I am very new to agar work with wild spores.
This will be my updates on how this project is going, and any advice would be greatly appreciated!!

What kind of media did you use? Idk what my prob is, i always have issues. Thats after a long career as a professional microbiologist too. My colonies form, just not in the way i want them. Tia!

@mycobabe just premade potato agar plate from amazon lol.
Figured they had a money back guarantee, might as well try to clean up these spores. I used to skip this step with wild collected, but dont have the time to find the perfectly rip cap anymore, also copelandia are almost impossible to get a clean wild print..

I am not subscribed to thise threads,.... I will def have to find those. And yes. I used PDA as well. They had the apoearance of a slightly mucoidal gram negative. Never would grow into rhizoids. I suspected contamination and was just going to pull the fungi oit, it never grew tho.

I'd think that most of that is mycelium but top right end blob looks like yeast or slime mold maybe? Transfer from the leading edge of myc at bottom of photo would be what I'd do.

Hopefully someone with more experience will be along...

(Disclaimer: My experience with agar work is very minimal!)

With wild prints I usually do one, add it to the collection and do another and use that for culture usually without cleaning needing to be done. The idea being that most contaminants drop on the first print and the second being much cleaner. I know it's frowned on by some, but I've spore swabbed straight to grain many times with no contamination. It's especially effective with fresh spores because they germinate quick. These are wild Psi. Cyanescens that were swabbed print to substrate with no contamination. Looks like yours are going great so far! 8798016b0eaefc12e8e8b52ae7f4b499.jpg

Thats what i used to do, make multispore prints tossing the first couple.

That cube's gonna be exciting! I've barely started with copes this year and have had nothing but problems on agar. Manure water in the agar mix really helps though. I'd say that center colony is most promising.
