agar work lions mane

hello i am new

 

i did some agar work for lions mane

 

some are from the mushroom itself. some are from agar to agar

 

please if anyone can help advise me which is not contaminated?

 

some are yellowish in color. some have wierd growth patterns

Welcome to Topia!

Most of those agar jars are very full of bacteria and/or yeast.

The last one looks less contaminated. I would transfer at least 1-2 more times to make sure there are no latent hitchhikers along before committing to LC, LI or grain from that agar plate.

Good luck, lions mane fruit bodies are very delicious and good for you!

i see so the yellow ones are yeast....

when you say the "last one" is less contaminated, did you mean the one on the lower right corner?

A couple of the brown jars look promising. Do some transfers from the thickest outer edges. Small pieces are best. Did you take the pictures inside a glovebox? I hope so.

no i did not take pictures in a glove box.

but i did open a HEPA filter first inside the room. and waited for 001 particulate before opening the jars and taking pics

i am thinking of buying an all american if my experiments work. 1. is there an accessory i can buy that can tell me when it reaches 20 psi instead of 15? 2. i have a few other experiments involving rice and coffee do i make a new post or keep posting on this one?

If you want to do anything outside of PF cakes, a PC is a must have. You also need to make a glove box to do work in. Those two items are the most important things to have. A hepa helps. Are you talking a flowhood or just a small hepa unit that cleans the rooms air? If the latter is the case, they do clean the air, but the fan also blows dust around. You would have much better luck running the small hepa for a half hour, shutting it off and then doing your work inside a SAB a half hour later.

5th try lions mane

 

first time using wheat. 1.5 hr in the pressure cooker. the bottom grains are stuck to the jar... :(

 

it was soaked for 24 hr first then transferred to 3 bottles.

looks like the agar to grain transfer is working

 

any advice when i should do grain to grain? and grain to spawn?

Shake those jars to break-up those solid masses forming in the bottom of the jar.

Allow the jar to recover for a day or two. If all of the kernels begin supporting mycelium you'll want to do g2g transfer at that time.

1 hour and 15min pressure cooker

 

1 container has almond nuts the bottle cracked at the bottom not sure why. the other bottles have no crack. it was soaked for more than 48 hr in 4c ref

 

1 container wheat soaked for more than 48 hours in 4c ref

 

1 oak chips was soaked for more than 48 hr in 4c ref

 

had to mix some of the wood chips into the other jars because it was full when i opened it

 

 

i did grain to grain and grain to spawn

notes on failure:

 

1. i tried to use old white rice. from agar to the used rice. it turned into a blob. smelled really bad

 

2. tried coffee grounds from starbux. agar to coffee grounds. it did not work either. the smell was not mushroomy even after a month

 

3. towel tek did not work

 

4. spraying from water bottle only created contamination (water was filtered water).

 

5. temperature is important especially in hot climates some  mushrooms cannot grow

started cooking 2nd batch today

 

2 bottles wheat. soaked less than 6 hr

 

1 bottle almond nut. soaked less than 6 hr.

 

- had problems with pressure cooker. pressure was venting from another hole. WD 40 fixed it. however i did not increase the cook time. it might cause contamination in the future. cook time is approx 1 hour 15min.

 

i am doing this because the bottle i used to transfer from grain to grain still has contents. i am afraid of contamination. might as well transfer everything.

 

started a 3rd  batch.

 

3 bottles of oak chips

 

 

 

note: i placed a circular wire at the bottom of the pressure cooker. it might prevent the bottles from cracking.

notes on pressure cooker:

 

1. aluminum foil cover over bottles to pressure cook is not needed

 

            - cotton over the hole on the bottle is enough

 

2. can use a large plastic tub filled with water. then place the pressure cooker on the water after it is cooked. it helps to lower the temperature fast.

 

 

3. placed a trivet at the bottom of the pressure cooker it might prevent the bottles from cracking

4. i had a problem with the pressure cooker. the steam was coming out of another hole. it turns out rust was blocking the center hole from making the steam escape. a few sprays of WD40 did the trick. after 10 minutes. the pressure cooker started working again

 

5. it looks like my cheap 5 liter pressure cooker has to be replaced due to the rusting of the central part.

 

         - i have used it less than 10 times.

         - it can only fit 3 glass bottles at a time

notes on clean room

 

1. buy a good quality HEPA filter.

                       - the good quality filters are not washable. you have to change the filter like a car aircon filter.

                       - it will have a particulate indicator. and will say how many particles are still in the air as it cleans it.

                       - mine can go 001 particulates. PMI 2.5 measurement

                       - i used something similar to this https://www.mi.com/global/air2/

 

2. i think a glove box is unecessary

                       - especially if you have a good HEPA filter

                       - note: i DO have a glove box. but i do not use it.

 

3. alchohol 70% is enough

 

4.  buy a butane torch

                     - it is way better than an alchohol lamp

                     - it is hotter and flame sterilizes the blade quickly

 

5. tissues are a must

 

6. a stainless knife is very useful

                     - do not keep it wet. it must be kept dry.

                    - there is little need to alchohol it. especially if you use the burner to sterilize it

                    - wrap with tissue when not in use (even if its stainless, it can still rust)

                   - use another knife or scissors to brush off the ash on the blade

4th batch. im too lazy to label a b c so i just advanced the date 1 or 2 days ahead


still having problems with the old pressure cooker. will replace


the all american will arrive october at the earliest.


the bottles are a little oily. i think its from the wire. it used to hold the used oil container...
