I haven't been using the liquid culture for awhile now. I've been experimenting with more low effort approaches. Hopefully it will lead to something worth a write up.

 

I'm kind of wishing I had saved some colonized grain of the Pan Peace River to do G2G. If the bulk substrate won't colonize properly then I will be back to square one with a multispore syringe. Oh well, eyes on the prize, eyes on the prize!

My pan experience was all about identifying the correct growth on agar. From there it was just keeping tent dialed in

My hope was to grow a batch of Pans from multispore and then inoculate some agar plates with inner cap tissue from the most promising looking fruits, and isolate from there.

All my MSS attempts were hot garbage, vendor syringes too. Needed a known print, but may have been just my lack of knowledge of culture selection. I may be way off base.

I am now working with agar to isolate the fastest growing strain and it is amazing how quickly the Pan Bisoporus grows! I inoculated an agar cup with a drop of MSS and within two days there was vigorous mycelial growth. I transferred a slice of agar that contained good growth to a new agar cup and by the end of the SAME DAY there was significant new mycelial growth. Today I am going to use a blunt needle to take a "biopsy" from a section of promising new growth to further isolate the strain on new agar and I will hopefully have a viable resilient strain very soon...

I agree bisporus have different mycelium, I think mine germinated just in two days, its way more thiner and transparent mycelium, but the growth is not much faster.

 

this is just horsemanure, hay, verm 2:1:1 , without any grains

In cans without grain, it really grows well,a lot of experience has shown that this is a good grain-free method.

I saw your fantastic grows without grains angam. btw are u doing multispore or clone?

 

here is my attempt at bisporus from spores. not great, most mushrooms are in this corner

Fruiting occurs separately in containers of hay-manure and cover from the ground.

Example:

I have a question about creating an effective fruiting chamber for Pans. I have a chamber based on the excellent DonShadow "Shadowbox" that has created good conditions for Cubensis and I am wondering if I can adapt it for Pans. The current setup has several inches of perlite in the bottom that has been watered to where the visible water level is about 2 inches from the bottom. Two 5" aquarium aeration stones that are beneath the perlite are fed air via a aquarium air pump. A CPU-type fan draws air in from outside through holes in the sides of the chamber that are just beneath the level of the perlite. The fan is set to be an exhaust fan and is on a programmable timer. There is a hygrometer inside the chamber with mostly reads "High," meaning the air is at or close to 100% humidity. That is, unless I program the timer so that the exhaust fan kicks in more often and for longer periods of time.

So dear forum member Pan experts, can I use this setup for fruiting Pans? I understand that Pans need more air exchange and more evaporation and I am curious what humidity level(s) I should be aiming for during the pinning and fruiting stages. Thanks very much.

Pace, my buddy JCM came up with a method that is similar to what you describe and it's pretty popular elsewhere on the internet.

 

Basically, it's a big tub filled with water and lots of air stones on the bottom. Your tray sits on a rack above the water. The lid has

holes drilled out like an SGFC. An aquarium heater at ~85F is used to maintain temps. No fans necessary. People are pulling

down some nice yields with this setup. It's just loud AF.

Hi Asura, Ha - I thought it would be more complicated than that, because I have read so many references to Pans needing evaporation of moisture in the casing in order to initiate pinning / fruiting. I found a write-up that DonShadow did of using the Shadowbox to fruit P. mexicana, which is what I am trying to grow now. He suggested an initial timing of the exhaust fan of 4 minutes every 4 hours to get that evap thing going on... So I have several shallow trays of P. mexicana with fresh casing that are now in the fruiting chamber hopefully on their way to pinning. However the grain spawn was colonized from a multispore inoculation so I dont know if this batch will be robust enough to fruit successfully or not. 

 

On another front, during the past few weeks I have been isolating a very fast growing strain of Pan Bisporus using agar and yesterday I added a few small wedges of promising mycelium-agar to a jar of liquid culture medium. Just a day later, and there is already very significant mycelium growth in the LC. Asura in a week of so when the LC is fully colonized I will use it in your mycobag tech for Pans.

If SWIM was going to start growing paneolus, how would I go about it? I'm assuming syringe -> grain -> substrate but what substrate, how h2o'd, are they better in monotub or some other arrangement, etc etc
