Cloning Pan Florida question

Hi guys...!

I'm going to clone one of my Pans today !
Using 3% peroxide and 2 or 3 parts of one stem.
They will be put in sterilized corn,that after full colonization..will be used the same as a normal colonized jar of corn.

Any comments or idea's ?

I failed the first time using a blender,so this will only be my second try !

why not try a liquid culture..?

I think that will not be suitable for Pans,I don't say it can't be done but if blending them kills them,then I think sucking live culture in a syringe will do the same thing...

And I think that I cannot use a liquid for spawning poo,..so I have to innoculate corn again.. this will take more time that I do not have !

But if you don't agree just say so !

LC worked well for "blue Helix" ..i think its just that pan myc is more sensitive to the presence of bacteria and does not recover from mixing n shaking...If your stems are meaty enough to get a clean interior section then both ways should work..I prefer the popcorn idea though as you can more easily see whats happening ...Then go grain to grain for expansion if ness....gluk..

Okay Golly ..I have put two pieces of a stem in peroxide for a short wile.
Then I cleaned it off, with distilled water very well...
I couldn't take the inside of the stem,because it was to small.

There I now in the glas along with the corn as you can see in the picture below.
If everything goes well,then I use this jar for spawning horse poo,and maybe take a couple of colonized corn pieces out, for further experiments !

Looks a bit wet Space...If your using just 3% H2O2 then there is a rather high probability of bacterial contam when taking an entire stem cross section...
If you have some more Pans to clone i would attempt to extract inner tissue with a pair of tweezers -just a tiny piece is all thats needed...

I have the Viets on there way,..I'll try it with them..there are a little thicker right ?

I don't care if this one failes,just a quick experiment to fill my time...

Yeah,the stems of Vietnamese are easily thick enough to split and extract.
The clone will take about 4 days to show new mycelial growth - they usually turn black first and look like they're dead B4 sprouting...
I've lost my fair share o clones which hopefully means - i've learned alot lol....Glukkkkk

Man, it would be hard to rip them small stems open and get some inner tissue. Some kind of precision biopsy tool would be nice.

chop a cross section then use a sterile needle to separate the strands

Never tried something as small as one of those, but with the bigger guys, letting them dry a little makes the tissue inside the stem shrink away from the exterior and become better differentiated and easier to remove.

I have to say, I've never seen one of these guys in person. Just how small are the stems? From the popcorn jars, they look very small, about 1/3 the diameter of a pencil.

Also, a small spoonful of dry verm will soak up any extra moisture on your popcorn.

Viets and Goliath have stems of 3mm+ on the best of em, which are splitable ...Seven says he peeled the "skin" off the cap on the smaller ones to get a chunk of meat but said it was a PITA...

is there a specific reasoning that you want to go directly to grain?

You don't have to use stem sections. You can always cut a sliver from inside a split cap.

BuckarooBanzai..that verm. thing is a good idea too,but I don't think I'll do that this time,..looks wet and it is a little bit,but don't forget it is a close-up picture !

Just a few minutes ago I went to have a look at my corn/clone jar...

I didn't expect to see anything yet,but was very suprized to see signs of growth on all two mushroom stems ! (picture)
Will keep you all updated on the speed of growth !

Well,..I do the same things with cubies,so why not ?

Caps may hold some spore that would get in the 'way' and germinate not ?
Do you think cloning a cap is better Dail8..,or just less difficult ?
