Found some fresh LM in the woods on a hike with my girl and the dogs the other day. My question is how do you take a tissue sample for agar?

Is it as simple as sliceā€™n open and taking from center.

Yes it can be. A lot of people do an H2O2 dunk but I never have. If you take it from a fresh exposed area you should be good. Core stuff not surface.

Don't slice the mushroom with a tool. 

Instead, rinse the fruit-body thoroughly and then tear the sample and grab tissue from the un-touched, previously un-exposed area revealed by the tearing of the sample. 

 

Cutting will introduce a contamination vector from the surface into the cut.
