Hey Topia!

so here is my first attempt at some culture plates.

I really dont know what i am looking at it. they seem good to go to me but I dont know how I should cut them up. Any suggestions?

Nice job, they look pretty! Unless you are isolating, you can go from there to grain/sub or LC.

Easier to clone a big specimen then put in plates upon plates for an isolate, IMHO

Still I wouldn't hesitate to grow those plates out and make some grain pints or quarts......that looks like something that needs put to a fruiting sub and grown out.....

 

I'd expand them first and then grow them out and put wedges to sterilized rye. Them ropes are worth growing out and experimenting with IMHO....

When I want info, I go to the old archives. Everything is way easier to find what you are looking for. Otherwise, I use google to pull up what I am looking for here.

They look perfect, I like to take a minuscule transfer, even just wipe a myc rhizo with your tool. Label both of them accordingly, and if that colony fruits like a mofo. Keep that plate as a master.

A flow hood with ample space to always have an open path to the filter and work in a natural and comfortable seated position is bloody priceless.....

 

I absolutely love this thing...grains and bags are coming up real soon.....but agar is great fun....

 

@sandman..... I got some nice stainless tubing and intentions.......I will have an agar plug tool....

So if I'm going from spore to agar (which is very unlikely) should I put a drop of a syringe in the center or streak it?

Spores from a syringe to agar would be a small drop (as small as you can get to drop from the needle).........

 

Spore print to agar can be done by flaking spores from the print onto the agar or preferably by using a flamed inoculation loop and streaking....

Good morning guys!

 

About to make my first agar. And I've come to this deductively, but I have to double check.

 

So, in the example of the pics above, the agar is innoculated UNDER the surface right?

 

But with a clone, that's impossible, so you just stick it on top?

And lastly... Does the agar cook IN the PC. Or do I cook it first?

No....the surface of the agar is the working plane for most purposes......I'm working with spores and transfers on antibiotic agar...MDYAA (malt dextrose yeast antibiotic agar)

 

Does a fungus eat down into the agar? I've definitely seen that too but for streaking, transfers, and clone purposes I'm just working on the surface myself.....

 

The fungus goes where it wants.....

 

The agar definitely gets cooked in the PC when you sterilize (what container are you intending on using)? I mix all my ingredients like I showed, 1000ml flask and mag stir hot plate....

 

You'll probably want/need to do some serious hand mixing on low heat in a pot and then transfer to your sterilization flask......

 

Make sure you're set before you start cooking things......

Too late. I'm about to learn the old fashioned way.

Trial and error.

 

i don't have a stirrer. I followed the mea Tek from the first link (maybe the second?) He doesn't cook it before he PCs. I've never worked with agar before, so I don't know it's expected consistency before it's heated. So... Science time.

 

I've already poured some. I also don't have a device that measures CC's or ml in small increments. The smallest I have starts at 50. Having said that, had some water measurement mishaps. I'm going with the theory that they'll solidify in PC. So we'll see how it goes.

It doesn't say specifically that Tek. But he does mention avoiding the pour.
