Is it that scary? I have medically sanitary conditions in my lab.

I'm not sure how you plan to keep spores and other nasties from landing on your plates (upside down jar lids) but you will soon realize how medically sanitary it really is. Just cover that plate/lid to keep from drying out and wait a few days. There is nothing scary about microbes if you understand how they get around.

How exactly did they not survive the PC?

When I make agar, I cook on the stovetop first, then pour into my jars. Keep the lids loose and cover in foil. Then stack in the PC (I generally try to keep them above the water in the PC by placing on top of other jars, not sure if it’s necessary). After the PC finishes, I let it cool slowly, I place towels over the top of the PC and let it be overnight to cool as slowly as possible. Then the next morning, pull the jars out quickly tightening the lids and store upside down if there’s condensation (typically there’s just a little condensation on the side of the jars, but not enough to hurt anything.)

And yes, it does set up if you PC it, then you can pour into jars.

The video looks ok, but why go to all the effort of pouring the agar in a glove box? It’s really a PITA in my opinion. If you follow a no-pour tek similar to the description I mentioned in my previous post, you can end up with sterile agar dishes without the glovebox. Also by following a no-pour tek, you don’t need to worry about the agar cooling too much while in the process of pouring into the individual jars.

So here's my quick and dirty agar tek.

Take a flask, liquor bottle, hot sauce bottle, whatever you have that caps.

Put in the flask 10mg of MEA, and 10mg of agar, tare the scale if you have. Now add 500mL boiling H20. Cap and shake like a crazy motherfucker, should be mixed good enough, might see some separation.

Now clamp that cap as a tight as possible, and then back off a bit so it barely vents. Now cover that top in foil.
PC @ 15psi for 40 minutes. From here you can pour into sterilized containers (15psi @ 20 mins), or if you want just pour this agar in your containers, seal 'em, and PC for 40 mins @15psi.

I can clarify anything you need, just at work and will check in time to time.
