Hey folks,

 

I'm super happy to say that it's time to do the thing again!

 

Fahtster was nice enough to send me a couple of Penis Envy swabs, and I put those to agar on 3/21.  The swab tips were broken so I put each of them right into a plate.  They started growing just a couple of days ago:

[I have a better camera that I'll start using, but I lost the power cord and I'll need to replace it.]

 

I took two wedges from each of these plates today and now I have the original two as well as four in the second generation.  I labeled them well enough that I'll know which one came from which one.  I want to keep track because I might end up testing for aggressive fruiting before I do an actual run.

 

The plan is to keep everything on a small scale, make a few jars of Golly's stealthier honey, maybe preserve a good strain, and make some spore swabs.

 

 

 

I also started this variety on agar:

Look at the color on that one!  It's from 2017 and I don't remember what it's called, but I'm pretty sure it's cubensis.  It's labeled "GH".  Does that ring a bell for anyone?

 

It hasn't germinated yet after a week, but since it's four years old I'm happy to let it take its sweet time.  Doesn't bother me to let it just sit there while I do other things.

Holy CRAP!  I went and searched Golden Halo here on the site and found this thread from last May:

Those are the exact same prints that were gifted to me!  The label on the Alacabenzi print I have wore off and I couldn't quite read it and I didn't know what it was either.  But that's definitely the one I have.  Now I know what that one is too!

 

What a crazy start to these projects.  Thanks jrh!

BTW, the glancing I did seems to indicate those may be tricky, so you might want to read up on them.

There are pictures out there of beautiful, actually golden spore prints.  But I don't know whether those genetics preserved or lost.  Seems like if they were around we'd be hearing about them frequently.

Yeah I'm sure the USPS equipment snapped them.  No harm done, though.  I think leaving the swab tips right in the dish was the way to go anyway.  There is a third plate where I just wiped one of the swab tips on the agar, and that plate isn't doing anything yet.  I read in another thread that someone else had the same issue.  Thanks again for the spores!

Update: Definitely running into some bacteria and pink-colored contamination in my PE plates, so I'll need to do more transfers this weekend:

 

The first two pictures are the original two plates. I labeled them "I" and "II".  Number II still looks clean to me, so I could take more wedges from it for plates, grain, or LC.  Maybe all three?  It's nice to have options.

 

 

I.  This one has some gnarly stuff going on in it:

I'm mis-using the Stamets "P System" to keep track of which plates came first and which are transfers from earlier plates.  I say mis-using because his "P1" would be the first pure culture, and even the cleanest of these has a little dot of bacteria in it.  Again, I'll let them grow until this weekend and transfer from the cleanest one or two:

 

These came from gnarly plate I and have more issues than the others:

These came from plate II and are better but still have a little bacteria. The plate on the left, you can see a good bit of bacteria along the outer edge.  The plate on the right just has one little tiny dot at about 2:30, but of course one little tiny dot is bad enough:

Nothing yet from the "Golden Halo" plates.  I still expect to wait for a while for them to do anything.  On the upside, I don't see any contamination in them.

Welp, I figured out a big vector for the bacteria I'm seeing: the old parafilm I have is splitting open along the edges of several of my plates, where the top and bottom half come together.  Those plates are basically open to outside air.  Derp.

 

I'll start double-wrapping them to see if I can keep from throwing the rest of the film away.  I think there's a lot left.

 

The good part is, it's not just because I'm doing shitty work!

Very interesting projects! Pulling up a chair.

I just finished moving and getting comfortable so I'll be getting all my projects moving again. I hope to see ya over there as well. But either way thanks for your contributions and I'll be keeping a close eye. :)

I took the three cubie varieties I have from 2017 and made spore water jars.  I used the lids that I just posted about here.  They are Golden Halo, Alacabenzi, and "SC".  I don't know what SC is yet.  Something Coast?

 

Unfortunately, the two GH petris that I tried to start already were both lost to my parafilm issue--bacteria sneaking in through holes in the parafilm.

 

Putting these old prints into sterile filtered water will hopefully help the spores "wake up" after sitting for so long.  What I'll probably do is let them sit for a bit in the water while I tend to my Penis Envy, then I'll test them on some PF jars.  If any of them are clean and viable I'll start with PF jars and clone to agar.

 

But, like I said, in the meantime I have to get my Penis Envy figured out.  Two days ago I transferred from dish "II" above to seven new plates.  Today, all but one are contaminated with bacteria or pink mold.  BUT, none of this contamination seems to have come in from the edge of the petris.  Hopefully, that means I solved that issue by double-wrapping with the parafilm.  Also, to be fair to Fahtster, I may have introduced these contaminants myself.

 

I'll keep transferring away from the contamination and working toward a clean culture.

Edit to add that I'm also pouring some more agar this afternoon.  The recipe I'm using is:

 

10g Telephone brand agar

10g Light malt extract

1g Activated carbon

1g Nutritional yeast

A pinch of brown rice flour, because I'll probably be growing everything out on BRF cakes

So here is that one surviving PE plate from my last post, just before I transferred tiny bits of mycelium this morning from the ends of the best looking rhizomorphs to five new plates.  Fingers crossed again!

I also worked with the spore water I made last week from my 2017 prints: Alacabenzi, Golden Halo/CRS, and "SC", my mystery variety. 

 

To those three I added another variety.  I had a jar of Redboy spore water stashed away.  The strain performs really well, even multispore.  I remember hearing that the source had worked with it for a long time.  I'm glad to have that one back.

 

I made the Redboy spore water in 2018.  I tested it about a year and a half ago, and was happy to find out that it was still good--100% colonization with no contamination.  I'm hoping it will be good again.  It's been kept refrigerated and undisturbed the whole time.

 

Anyway, I'm testing each of the four out to see what I'm starting with.  I squirted maybe a quarter CC of the spore water into a petri dish each, and also made one or two PF jars with each variety.

 

Onward!

Sidestreet, this is looking really promising :) I'll have a bunch of different strains fruiting this next month and I'll send you some strains if you like. I'm planning on doing a big giveaway :)

I've got four strains that are germ'd and being transferred tonight and then four more strains that are going to be streaked.

Alright, it's looking good for my PE cultures.  I have three apparently clean dishes:

Since the plan is to test for a good fruiting strain for cakes, I'm thinking I'll take multiple wedges to LC so I still have a polyculture.  Actually, I might do one LC with the best looking rhizomorphs and another with multiple wedges.  With that LC, I'll probably do a small run of cakes to find a cloning candidate or two.
