Should I store it in refrigeration once it’s grown a little bit? Or do I let it take over all the available agar surface and then some?

How much do you let a culture slant grow?

It will continue to grow slowly after placing it in the fridge, so in theory if the entire surface isn’t colonized it doesn’t matter much unless you plan to use it soon. I suggest you let it grow out completely for the sake of ensuring there is no contamination present before long term storage. Imagine going to grab your slant in six months only to discover that it was contaminated from the beginning… and now you don’t have another backup.

Always let your agar slant colonize completely. I will allow it to grow down between the agar and glass a little before placing into cold storage.
