2 Years Out, 4CCs In!

Hello Mycoropia its good to be back,

2 years out, life doing its thing, but the supplies are out the fridge and we are giving it another go!

B+ MS, 4no 1 quart grain jars, soak and simmer WBS, 15psi for 90 mins, cooled in PC and noc'd the day after, 1cc in each.

Its been 3 weeks and 2 days and my jars have colonised 15%
Im happy the spores are still active and doing the biz but its not the characteristics im looking for, ive been well advised they may be a hidden contam slowing things down and to give them a shake and monitor recovery. That is what i have done and updates are pending.

I also recognise that MS > grain, isnt best practice just trying my luck and being lazy, i have had agar mixed and PC'd and stored in the fridge during this whole lay off. I PC'd again at 15psi for 30 minutes, allowed to cool and started off some spores to agar. Updates pending, its been 2 days.

Thought i would share, others starting off old spores may be able to find this some what useful.

Keep in mind that 1cc is a small shot (slower progress), I regularly shoot 2-3ccs. in quart jars.   That being said, a good shake like you are about to do will speed up the process.  If I do a small shot with what's left in the syringe, I'll do 2 shakes to speed it along, or a pint jar to start with.  Always options.  Good news is... you have healthy mycelium.   :thumbs_up:

Hey Mush! Thanks for dialing in. Appreciate it. Thats good to know ill keep that in mind. Ive always been under the impression more than 1cc could cause problems. Too much moisture. That probably stems from the PFtek method i was following when i first started out.
But yes! Healthy mycelium! Hoping to see some growth in the agar over the next few days aswel. Updates pending.

Hello people!

We have some activity going on in the agar cups but not what i was hoping for. I overshot the first cup and had far too much liquid on the surface of the agar so was expecting something bad from that. The other i managed to drop a few drops into the centre. However both cups are showing something funky.

At the same time while the agar cups were out and the SAB was set up i thought id put some JEDI MS to agar and those 2 cups are just starting to show tiny white dots with clear mycelium reaching out. All cups were poured at the same time.

Please take a look and share your thoughts!

PF Tek is a different beast for sure (1cc per hole or whatever), I've seen folks literally pour LC (like a G2G transfer) in a quart jar of grain and mix it and it's fine (you can do an LC straight from a plate, or Grain LC).  With grain jars, wet grains are more of a problem than a LC (then yes, at that point... added moisture to wet grains is no bueno).  Many ways to get to the end result.  Good Luck with your projects, hope to congratulate you at the finish line.   :meditate:

I have some Mazatapec LC in the fridge but its 2 years old. I have some Lions Mane LC also. Trying my best to stick to one project at a time but i will put them to agar in the future to test their viability.
As for this project the shaken grain jars are recovering well and the JMF is growing nicely on the agar. Still unsure about those B+ agar cups though??
@MushLuvR what are your thoughts? I can try and get a more clear image for you if those ones are no good?

Very slow but that is the price of going MS > grain

Agar cups showing growth,
JMF nice and clean with signs of rhizo forming! How long should i grow these out before transferring?

B+ agar cups are looking suspicious. Im torn between tossing them and starting again MS > agar or wether to transfer one of those islands to another plate?

Grain jars are coming along nicely. Its took a while but we are talking 2 year old MS straight to grain so getting this far is good to me. Another week or so and they may be ready for spawning.

I decided to transfer from the B+ cup one of the rhizomorphic looking islands into another agar cup. One wedge dropped into the centre and this is the growth i am getting after 11 days. It doesnt look likecubensis mycelium to me it is reminding me of lions mane. What do you guys think?

Any advice is greatly appreciated. Thanks in advance.
