Marijuana Research Offers New Hope For Male Birth Control Pill

The male birth control pill has lingered for years tantalizingly just out of reach, in the realm where rumor meets science. Recently developed hormonal and mechanical contraceptives never found an audience, serving only to highlight the absence of a male pill. Now, an examination of how smoking pot lowers fertility may make the male pill more than a persistent rumor.
Writing in last week's issue of the journal Cell, University of California, San Francisco, researcher Yuriy Kirichok revealed a new link between bong hits, a protein called Hv1, and the ability of sperm to swim. Hv1 activates in alkaline environments, like the vagina. Kirichok's study showed that endocannabinoid anandamide, a chemical found in the kind bud, raises the testes' pH. In that raised-pH environment, sperm start swimming too early, and get too tired to reach the egg once they actually exit the body.
A pill that contains anandamide, or in some other fashion alters the pH in testicles, could make anyone's sperm as lazy and apathetic as the spliff-ripping burnouts gaining that sterility the hard way.
However, before every tosses out their condoms in favor of some chronic, it should be noted that an Hv1 pill can't hit the market until some scientists explore some potential side effects. Specifically, Hv1 protein acts across the body in a range of different ways, not just in sperm. By changing the pH environment for Hv1 protein in general, bodily functions may alter in unforeseen ways. So while this is definitely a positive step towards creating the long-desired male birth control pill, it may not be available in stores until after Gilliam finally finishes his Don Quixote movie.

The predictable swimming sperm featured in health class videos are rather uncharacteristic portraits of the life of these sex cells.

Spermatozoa actually spend most of their time resting up in the male reproductive tract so they can make a dash to the egg once the opportunity arises. It has long been known that the little swimmers get their cue to start moving from a big change in pH levels, but a new study explains just how that shift occurs: a specific molecule in the sperm cells' tails releases protons when the time is right.

"The concentration of protons is extremely high at all times while the sperm are in the male reproductive tract, which makes the intracellular sperm environment acidic and inhibits the activity of the sperm cell," Yuriy Kirichok, of the University of California, San Francisco, said in a prepared statement. So releasing many of those protons from inside of the sperm cells lowers the internal acidity, a cue to the sperm cells to start flipping their flagella.

But how does that proton release happen? Previously, researchers had been unsure because they were not able to measure the membrane conductance on human sperm. But by using a whole-cell patch-clamp method to measure electrophysiological changes, Kirichok and his team were able to pinpoint the specific molecule—called Hv1—that opens up proton channels on the sperm cell membranes to release the extra protons. "The Hv1 channel, when opened, can allow protons to exit, and activates a cascade of biochemical reactions that cause the spermatozoa to move, mature and prepare to fertilize the egg," said Kirichok. This channel is signaled by increased pH and decreased zinc—both relative characteristics of the female reproductive tract compared to that of the male.

Another change in the environment that seems to spur on the sperm is an endocannabinoid that is common in both male and female reproductive tracts. The role of the endocannabinoid system also raises questions about the reported link between marijuana, which activates cannabinoid receptors, and male infertility. "Marijuana likely activates sperm prematurely, leaving them burnt out in a matter of hours," said Kirichok, who is a co-author of the new study, published online February 4 in the journal Cell.

A better understanding of sperm cells' swimming tactics might open the door to different methods of contraception. "You can imagine now that we know the molecule responsible we could block it to prevent activation and fertilization," Kirichok noted. Conversely, those spermatozoa needing a boost might some day get a little help with their Hv1 channels.

Cannabis sativa (also known as marijuana) has been cultivated by man for more than 5,000 years. However, there was a rise in its use in the 20th century for recreational, religious or spiritual, and medicinal purposes.

The main psychoactive constituent of cannabis, whose structure was identified in the 1960's, is Delta9-tetrahydrocannabinol. On the other hand, the discovery of cannabinoid receptors and their endogenous agonists took place only very recently.

In fact, the first cannabinoid receptor (CB1) was cloned in 1990, followed 3 years later by the characterization of a second cannabinoid receptor (CB2). Since the 19th century, the use of cannabis has been reported to stimulate appetite and increase the consumption of sweet and tasty food, sometimes resulting in significant weight gain.

The recent description of the endocannabinoid system, not only in the central nervous system but also in peripheral tissues, points to its involvement in the regulation of appetite, food intake and energy metabolism. Consequently, the pharmacological modulation of the over-activity of this system could be useful in the treatment of the metabolic syndrome.

Conclusions: The endocannabinoid system has important physiological functions not only in the central nervous system but also in peripheral tissues.

The activation of central CB1 receptors, particularly in hypothalamic nuclei and in the limbic system, is involved in the regulation of feeding behavior, and especially in the control of the intake of palatable food. In the periphery, cannabinoid receptors are present in adipocytes, skeletal muscle, gastrointestinal tract and liver, modulating energy metabolism.

The active ingredient in marijuana cuts tumor growth in common lung cancer in half and significantly reduces the ability of the cancer to spread, say researchers at Harvard University who tested the chemical in both lab and mouse studies.

They say this is the first set of experiments to show that the compound, Delta-tetrahydrocannabinol (THC), inhibits EGF-induced growth and migration in epidermal growth factor receptor (EGFR) expressing non-small cell lung cancer cell lines. Lung cancers that over-express EGFR are usually highly aggressive and resistant to chemotherapy.

THC that targets cannabinoid receptors CB1 and CB2 is similar in function to endocannabinoids, which are cannabinoids that are naturally produced in the body and activate these receptors. The researchers suggest that THC or other designer agents that activate these receptors might be used in a targeted fashion to treat lung cancer.

We discovered that cannabidiol (CBD), a non-psychotropic compound from the plant Cannabis sativa, can inhibit the process of breast cancer cells that allow them to grow and spread (metastasis). CBD can also inhibit breast cancer metastasis in a mouse model.

The research carried out in our CBCRP proposal demonstrated that CBD is a novel inhibitor of a gene whose activity is intimately linked to the aggressiveness of human breast cancers; this gene has been termed Id-1. Notably, our findings also indicated that Id-1 was a key gene whose expression needed to be reduced in order for CBD to inhibit aggressive breast cancer.

One of the most significant high risk components of the initial application was to determine if CBD had appreciable efficacy against breast cancer in vivo (i.e., animal models). This high risk component was not pursued, since an independent group showed CBD was able to inhibit metastasis of MDA-MB231 cells to the lung of nude mice. Building of the previous findings, we made small structural changes to CBD that are expected to produce drugs that are much more active than CBD at inhibiting Id-1 and corresponding aggressive breast cancers. CBD has a low toxicity profile. An anticancer agent with a low toxicity profile that can both inhibit cancer cell growth and metastasis would be extremely valuable clinically.

Understanding the mechanisms behind the anticancer activity of CBD may also lead to the discovery of new biological targets for the development of diagnostic tools and additional therapies for the treatment of cancer. In this project we found portions of the CBD structure essential to its biological activity for breast cancer cell growth inhibition. We are in the process of filing a patent on these discoveries. In addition, we studies the moleculr mechanisms that underlie CBD activity, and found that sustained upregulation of Erk (extracellular signal-regulated kinases, a type of protein kinase intracellular signaling molecules) is key to the ability of CBD to regulate the metastasis-specific inhibition of the Id-1 transcription factor.

Antioxidant activity has been reported to be a general property of the phenolic components of marijuana. Unlike Δ9-THC, cannabidiol can be administered at relatively high doses without undesired toxic or psychological effects.1 Cannabidiol at a concentration of 10 µM was neuroprotective against both excitatory neurotransmitter (glutamate) and oxidant (hydroperoxide) induced neurotoxicity.

There's a bestseller titled The Botany Of Desire: A Plant's-Eye View Of The World, by Michael Pollen that includes a long section on marijuana and how pot makes us high. According to him, the feeling of being high results from thc's mimicry of the our endogenous cannabinoids. Those endocannabinoids act as sensory filters that allow us to ignore, (forget), sights, sounds, and other sensory data that would otherwise overwhelm us and prevent us from focusing our energies on any given task. Memory, he says, is the enemy of wonder, and by changing how memory works, thc leaves us in a state of wonder. I had assumed that the temporary short circuiting of short term memory I get when I'm high was a secondary consequence of thc's action, but maybe the way thc changes how we create and store memorys is much more primary to the feeling of being high than I though it was.

Another good question is why the cannabis plant would have evolved these very specific chemical compounds that just happen to react with our human endocannabinoid system.

Thanks for the correction Hyphaenation!!

One of the more interesting chapters (to me at least) relates to the appetite modulation of endocannabinoids-I have summarized as best I can in my own words so I hope it is not too longwinded or boring and that I haven't diluted the technical content too much. It interests me that what we eat (since we would have eaten cannabis before we smoked it) can alter our behavior and direct what we eat in the future. How many of our behaviors would be altered by the plants around us that we should be eating? I suppose it even leads to the question has our diet made us different than our ancestors if we really are what we eat?

Anyway I digress:

Basically this chapter explains the rise in obesity levels as a result of insufficient feedback and suppression to prevent people over eating. The idea with most of the bodies signalling pathways is that they will regulate their own activity and a signal will, in one way or another, give rise to another signal to reduce its expression or activity. For evolutionary reasons this control mechanism has not developed for our response to food/ energy intake- it was never a problem for our ancestors I suppose rather there was usually not enough food.

We are as such "biologically programmed for gluttony" which challenges the idea that obese people are greedy or lack control rather they are designed to be obese in a world where there shouldn't be enough food to be obese! It appears that even nature wasn't prepared for a species with too much food. THC appears to play a part in this mechanism (here I am reminded of the simpsons episode where the marinol drug comes with the warning that it may make things seem more edible than they really are) since we eat to satisfy "perceived" cravings (since we are usually well nourished) rather than "real" cravings resulting from malnutrition from our bodies.

Obviously, as THC has a receptor there is an endogenous system where it plays a physiological role and its mimicry of the endocannabinoid system lends itself to study. In the absence of THC the bodies own drug/ transmitter anandamide fulfils this role and increases appetite (as does the endogenous noladin ether) by acting on the CB1 receptor subgroup.The CB2 receptors do not seem to affect appetite in the animal studies. Conversely antagonists (rimonabant) have been shown to prevent this mechanism. The CB1 agonists activate this pathway to improve the sensations associated with food intake- which roughly translates to the munchies and since it is amplifying pre-existing sensations. It makes the sweet taste sweeter so to speak so that we are more likely to consume reward food as it is more desirable from an evolutionary point of view. In addition it also makes the sweet appear sweeter as the effects of the alteration of the perception of food intake begins prior to tasting and the agonism of the CB1 receptors increases the drive to eat. Perhaps this in the past was a drive to survive if hunger wasn't pressing but motivation to hunt or gather was necessary?

It has also been shown that anandamide and THC contribute to dopamine release which may be responsible for the reward process. There is obviously a lot to explore with this if the reward systems and the endocannabinod systems are interlinked like this and indeed the opioid receptors even come into play. Nalaxone has been shown to inhibit the hyperphagic activity of THC which could prove beneficial in the form of properly balanced medications for both pain and holistically to build appetite :)


I would be very interested to see if the role of the endocannabinoids have been examined in the case of anorexia or bulimia to see if there isn't some antagonism going on in there.

Anyway I have probably hogged enough server space as it is!!
