hey yall... I haven't posted anything in awhile and i'm bored so i thought i'd show ya what i'm up to.. nothing really differnet here..
this is the same isolate ive had for over a year.. but everytime i use it (which is about once a month) it's only on it's second 'round' of use.. what i did was make 24 'faht jars' (i don't what else to call them) with about 1/2 inch of corn in the bottom and knocked those up with the isolate..
then, just before they colonize the corn, I remove all the tape from the tyvek (except for the tape covering the innoc. hole) that would normally keep the jars from drying out... this allows them to dry out.
i do this right before the jars are colonized so that the mycelium doesn't go into fruiting stage. this helps keep sessenence (sp?) at bay. mycelium can be reanimated so these dried out 1/2 pints last forever..
drying them out also helps keep bacteria down i believe. it's also easier to 'harvest' more myc. when they are dry because the myc. becomes brittle and breaks into very very small particles which is nice. :) I harvest the myc. by injecting plain sterilized water thru the tyvek via syringe... then let it sit for a few minutes and shake it up. then suck the myc. laden water back up. then shoot that into a covered sterilized 1/2 pint filled with water that i suck up with how many ever syringes i need for the task at hand. :) the last two pictures are of what i was just talking about.. the dried up myc. corn ready for harvest at will. :) then when i use the faht jar.. i just replace it with another one... so when it gets used again in another year it'll only be on it's third 'round'.
also, a dry ounce was already picked from this bin prior to taking the pictures..

i'm missing something-
that's not a thin layer of popcorn fruiting,
right ?
you're using the pcorn to spawn with yes ?
or ????

i re edited it now so it should read better... i use that dried up corn to harvest the myc. from.. so yes, i use it as spawn in a way :) thanks for the kind words.

so the cakes are pf style ?
nice isolate i'd say.
from which 'strain' was the isolate derived ?

hehehe... basically i knock up some corn in 1/2 pints, let them dry out, and harvest the mycelium so that i can make syringes with that myc. to knock up pf jars. :) i don't fruit out the corn. I use the corn as kind of a 'crumbling agar' to grow more myc. that i can knock up pf jars with or whatever the growing medium may be. these are double stacked 1/2 pints.. so there are 24 1/2 pints in that bin.. it's good ole texas. :)

how fast does the dried tissue re-animate
and how fast, once revived, does it grow,
i.e. how does it compare to either live cloned tissue
or even multi-spore
time-wise ?

it takes about two days to see visable growth once re-animated... I think it's a little bit slower than live tissue but not much definately beats out spore innoc. :) live clone tissue is pretty instant i would think... once it hits the substrate it's on i'm guessin.. but couple days isn't all that bad. :)

how long is the dried tissue viable,
able to re-animate ?
what's the longest you're heard of between drying and use ?

well.. I'm going on over a year for mine... haven't really heard a whole lot.. someone mentioned that mushroom tissue could be re animated... i think it was stephan peele.. it was awhile ago.. I was already using the 'faht jars' but i was 'harvesting' the myc. when it was still growing.. then i got the idea to dry them out and it actually worked lol.. then everything came from that.. i.e. making 24 'faht jars' to prolong the substrain use without damaging the strain from constant cloning and getting mutations... eventually the strain will mutate but it will take quite awhile for that to happen i think.. years + :)

and 24 was kind of excessive... i probably would have been happy with 12 faht jars... oh well :)

i store them at room temp.. but i was thinking that the fridge would be better.. don't know about the freezer tho.. there still is some water in the corn and myc. i would think.. so the frozen water crystals may ruin the myc. cells. I guess i could try it once. :) i haven't been the greatest at storing my jars.. didn't really put all that much thought into it. I just keep em in my closet. lol i haven't seen any ill effects tho, so maybe thats why i haven't paid all that much attention.. but i would definately recommend the fridge and wrap jar in tin foil to keep out light. I've been thinking about doing a new strain or finding the mazzies again.. really liked that strain.. so all the jars i made probably aren't going to get used. tis life. :)

Nice lookin thread you have goning here. Are those pics of an Isolate that you produced through your fahhty isolation tek? Or an isolate worked from agar? I am, currently useing your tek with some wild Florida Cope strain and I hope they look as good as your Tex isolate. Nice work

I tried the fahtty iso tek. A little bit of corn was put into the bottom of the jar, with a couple of screws. Once fully colonized i squirt water in a shake it about, then draw the water back out.
This usually makes for nice myc syringes. Somone suggested to me sucking a bit of karo out of a sealed jar (with airport) into the syringe as well.

Great info and nice isolation.
