47,liquid to white rice with horse poo and colonizing
48,those are dead
49,the others dunked after full colonization

back to clone No 5,the mushroom from which mycelia grew produced mushrooms again,so some of those were taken and placed on fresh PDA,
from this comes the first post in this thread,
it is a clone from a clone straight from the petri!

well,that is about all,
have saved little liquids of every isolate and clone and hope in the future will have the opportunity to test them :)

your pda is so clear.,.,,what recipe are you using and procedure for autoclaving? I'm using MEA and tried pda but both times it always clouds a bit on the bottom.

i like the way you put 4 isolates onto one plate , thats saves money in the long run plus its more effective imo . i do the exact same thing . nice work up .

do not remember now how much PDA powder for a litter of water it was,will check and let you know,
the autoclaving is supposed to be for 15 minutes but i prefer 10 to 13 minutes with a risk ofcourse,

and if you want clear, pour the agar very thin in the petridish

no,haven't tryed anything different than what you see,besides this is already too much for a lone searcher :)

pouring agar to thin causes it to stick to the sides of jars when trasferd to grain . it is a good way to pour them when your doing agar to agar tranfers . pda is a good recipe for everybody cause all the ingriedents or easy to get .
i like 300gm of taters in 1000ml of h2o boiled for a hour thenstrained threw cheese cloth . added to 20 gm agar and 10 gm dextrose . makes for a beautifull clear agar everytime . also i go for 40-45 min pc at 15pci anything less seems to have a 10-20% contam rate . anything over 45 min at 15pci causes the media sugars to carmalize .

:) second flush,
after the first flush the block was washed under running water,drained and put back in the fruiting chaimber,
no dunking,no caseing and not bad for a second flush also :):)
