PE Grow Log: Agar Isolate to Rye Berries -> G2G -> Monotub

Earlier today, I noticed I had a quart of PE on a shelf, fully colonized, since mid September.  It was inoculated with an agar wedge containing a PE isolate.

 

As you can see in the pic - it's very dehydrated and bruised blue from me trying to beat it/break it up a bit for the grain to gran transfer.

I had 3 empty quart jars available; so I prepped them with Rye Grass Seed (soaked RGS/gypsum/diluted coffee/boiling water for ~ 5 hours) and PC'd at 15psi, for 90 minutes.

As of right now, the jars are cooling, then I'll G2G in a SAB.

 

Once fully colonized,  I intend to spawn to bulk - monotub.

Awesome :-) love seeing pe grows. Heard rgs Is tricky to prep right though

I've used it on several grows (cubes, tampanensis and galindoi) and it never really seemed to be any more difficult than prepping any other grain(?)

Very nice. Haven't tried it yet just what I've heard but nice to hear it's fairly simple

Yeah, check it out - many growers swear by it.  I loved using rye berries, then I read about RGS and found 5lbs of it for $7 - which was much cheaper and easier to find than rye berries

I had a jar do something similar I shook the hell out of it and what not and the next day it looked like it was contaminated it never even crossed my mind that it might have just been bruised.

Yeah, usually the sign of contams after shaking grains is the mycelium fails to recover/recolonize.

 

This jar had been fully colonized for a couple months, so I'm nearly certain that it's just bruising from me banging the jar, exacerbated by the dehydration of the sub.

 

Anyway, I'm going to do the G2G transfer tonight.

Just G2G'd - which sucked, since the mycelium had consolidated to such a point that it was breaking off in large chunks (which is why G2G is recommended at exactly 100% colonization).  I used a disposable scalpel that I usually use for agar.

 

My SAB which isn't fancy, but has been pretty reliable.

I'm just curious if I can get a good grow from what are essentially spare supplies and 1 old quart jar.

 

We shall see....

Ah heck guys.  I use partically col jars of all sorts all the time.   Uncle G on soap box, stop letting the myc decide the time table of the grow and make things happen.  

 

Mycomattie try a sterilized fork next time.   You can  sterilize it a couple ways I prefer wrapping mine in tin foil and pcing them.   Or if you forget pouring iso over the end of the fork and waiting for it to dry in the gb works.  Flame works but you still end up waiting.  In a sterile environment such as a gb jars dont have to be fully col.    

 

Many times I use a 50% col jar scarpe down to the un-col part put the lid back on and let it fully col and use the rest for something.   

 

Not trying to be pushy just trying to help people have an "ah ha moment".

Funny you should bring it up!  I DID wrap a fork in foil and PC'd with the jars.  For whatever reason, the next day, when I went to G2G; my girlfriend must have shifted things around and it was kind of hidden, because when I went to transfer, it totally slipped my mind that I had the fork.

 

I agree with "not quite 100%" colonized but remove uncolonized grains and spawn.  I do that all the time too - I just don't bring it up since many consider it "bad practice."  Though it never really caused a problem for me.

Ah the joys of home mycology - patience...

 

This PE was originally from a MS syringe and it's overall colonization speed is SLOW - even when the rhizomorphic sectors were isolated.
