didn't really want to create a grow log, maybe just some random pics of what i have going on.
these are in no particular order as i have been transferring a bit. you may be able to see which transfers the plates are currently on by the "x #"

Hey pulling up a chair.
Been watching the agar threads for a while.
My first plates were well over a year ago. They did terrible and it took me a year to get back to it again i just couldnt give the plates the time and patience they needed to proliferate.

I am very excited about this Whitethumb!!!!! I am trying to soak up anything agar and Pan related, very excited to see your work, would love it if you also follow this through to actually growing and fruiting the Pans. This is priceless knowledge and I am glad to be a part of this group and this thread...

Dumb question, but if I want to get something growing on Agar, how many Petri dish we should I use to make sure I get the best strain and its not contam? I do all the work in my SAB and wrap them in plastic as shown by Whitethumb and then wait for them to grow. How much do you use from a syringe per plate and how do you get them all so perfectly centered?

Sorry, these are 101 questions, not 501.

no worries. as far as how many petri's you need really all depends. are you getting your spores from a trusted and clean source? if so you won't need many because they should be somewhat clean so you wouldn't have to clean it up much. you're really just looking at how many do you think you'll need to get the growth you want and then fruit it. i have never used a syringe in petri's, i usually avoid them if at all possible. if i did use a spore syringe i would put 1 drop in the center of probably 2 or 3 different plates. all the pictures you see have at least one transfer so when i take a agar wedge from my master plate, i put that wedge in the center of the receiving plate. hope this helps

So these plates have already been selected from the initial plates you used, correct? And you only have to do one transfer since you know what you are doing so well??? Or is because everything was clean?

1. So I grow out 3 plates of a Pan, then select based on advice from the group, how long should this take?

2. After I grow the first plates, do I select the best strain and put it on one plate or 3 more? And continue this process as long as needed, which is at least 1 transfer, correct?

1. pretty much. it all depends on what exactly your goals are. are you just wanting to make sure you have a clean culture? or do you want to isolate for desired growth? if clean it's what you want that can take 2 or 3 weeks depending on how long it takes to germinate. isolating can take a month or 2 depending on how soon you can isolate your desired growth.

@microbe77 taught me to put a divot in the center of the agar for one drop of spore solution when using a syringe. Getting one drop out takes enormous amounts of patience IME.

 

 

I've been streaking spores directly on the surface copying @peace.
