There is some agar blended in and can get chunky. The needle is stainless steel, 3.5 inches long and a 13 gauge beveled tip. If anyone wants to start using this kind of equipment the tip of the needle should not touch anything. If it is pushed into a glass jar the chances of bending the point are great making it difficult to use on the next project. Glass syringes and stainless needles can be autoclaved and reusable.

last year I was able to revive a Azur culture that was blended and stored at room temp from 2012. It made it to a fruiting tote/tub then outdoors. Weather went from summer to almost winter here so all we got was a few pins. Just enough to start a clone and begin to acclimate and develop a northeast strain of azur. being a small amount of tissue to work with, my choice for agar was PDYA plus H202.Upon closer examination there was a tremendous amount of mycelium growing from the pins overnight. Its not to sure about the H202 agar yet but with time it should start to expand. I also found a couple of transparent worms living in the body of the pin. I will try to get a phase contrast micrograph of them if possible real soon..

Mycelium growth has been extremely aggressive from this tiny pin, that small larva has been covered plus better to not open the plate anyway. Plate choice was a 150mm glass Pyrex petri and sealed with laboratory tape. The close up was taken through the glass and hard to see but I circled a colony of bacteria that came with the transfer despite using hydrogen peroxide. There Is also a very aggressive mycelium growth on the agar already, I'm kind of nerves about it so will have to keep a eye on that.....
